چکیده

APOBEC3B (A3B) is a cytosine deaminase that converts to uracil in single-stranded DNA. Cytosine-to-thymine and cytosine-to-guanine base substitution mutations trinucleotide motifs (APOBEC mutational signatures) were found various cancers including lymphoid hematological malignancies such as multiple myeloma A3B has been shown be an enzymatic source of those cancers. Although the importance being increasingly recognized, it unclear how expression regulated cancer cells well normal cells. To answer these fundamental questions, we analyzed 1276 primary using single-cell RNA-sequencing (scRNA-seq) was preferentially expressed at G2/M phase, sharp contrast patterns other APOBEC3 genes. Consistently, demonstrated protein phase by cell sorting. We also blood expressing enriched G2/M-phase analyzing scRNA-seq data from 86,493 bone marrow mononuclear Furthermore, revealed mainly plasma cells, CD10+ B erythroid but not granulocyte-macrophage progenitors. profiling may contribute understanding defense mechanism against viruses, partially explain bias APOBEC signatures myeloid malignancies. This study identified cellular which highly expressed, help reveal mechanisms behind carcinogenesis heterogeneity, biological functions

The Oenococcus oeni clpX homologue is a heat shock gene preferentially expressed in exponential growth phase.

Using degenerated primers from conserved regions of previously studied clpX gene products, we cloned the clpX gene of the malolactic bacterium Oenococcus oeni. The clpX gene was sequenced, and the deduced protein of 413 amino acids (predicted molecular mass of 45,650 Da) was highly similar to previously analyzed clpX gene products from other organisms. An open reading frame located upstream of ...

Sodium butyrate blocks HeLa cells preferentially in early G1 phase of the cell cycle.

The effects of sodium butyrate on the cell-cycle transverse of HeLa cells was re-examined using the kinetic and the premature chromosome condensation methods of cell-cycle analysis. The results of this study indicate that the sodium butyrate effects are dose-dependent and cell-cycle phase-specific. Cells in the early G1 period are the most sensitive to the inhibitory effects of the drug. When H...

The hyaluronate receptor is preferentially expressed on proliferating epithelial cells

In the present study, we have examined the distribution of the hyaluronate receptor as well as hyaluronate itself in a variety of adult tissues. The hyaluronate receptor was localized with a monoclonal antibody, termed K-3, while hyaluronate was localized using proteolytic fragments of cartilage proteoglycan. Staining with the K-3 monoclonal antibody revealed that the hyaluronate receptor was p...

Celecoxib toxicity is cell cycle phase specific.

Celecoxib inhibits proliferation and induces apoptosis in human tumors, but the molecular mechanisms for these processes are poorly understood. In this study, we evaluated the ability of celecoxib to induce toxicity in head and neck squamous cell carcinomas (HNSCC) and explored the relationships between celecoxib-induced cell cycle inhibition and toxicity in HNSCC. Celecoxib inhibited the proli...

B cell chemoattractant CXCL13 is preferentially expressed by human Th17 cell clones.

Th 17 cells represent a novel subset of CD4+ T cells that have a protective effect against extracellular microbes, while they are also responsible for autoimmune disorders in mice. However, the protein expression profile of Th17 cells remains to be clarified. In this study, we report an effective method to establish human allo-reactive Th17 cell clones and demonstrate that human Th17, but not T...
